plasmodium falciparum: adhesion phenotype of infected erythrocytes using classical and mini-column cytoadherence techniques.

چکیده
background: cytoadherence of plasmodium falciparum - infected erythrocytes to host cells is an im-portant trait for parasite survival and has a major role in pathology of malaria disease. infections with p. falciparum usually consist of several subpopulations of parasites with different adhesive prop-erties. this study aimed to compare relative sizes of various binding subpopulations of different p. falciparum isolates. it also investigated the adhesive phenotype of a laboratory p. falciparum line, a4, using different binding techniques. methods : seven different p. falciparum isolates (itg, a4, 3d7 and four field isolates) were cultivated to late trophozoite and schizont and then cytoadherence to cell differentiation 36 (cd36), intercellu-lar cell adhesion molecule-1 (icam-1), and vascular cell adhesion molecule (v-cam) and e-selectin were examined. the relative binding sizes of parasite subpopulations to human receptors were measured by mini-column cytoadherence method. the adhesion phenotype of p. falciparum -a4 line was evaluated by in vitro static, flow-based and mini-column binding assays. results: the relative binding size of itg, a4 and 3d7 clones to a column made with cho/icam-1 was 68%, 54% and 0%, respectively. the relative binding sizes of these lines to cho/cd36 were 59.7%, 28.7% and 0%, respectively. different field isolates had variable sizes of respective cd36 and icam1-binding subpopulations. a4 line had five different subpopulations each with different binding sizes. conclusion: this study provided further evidence that p. falciparum isolates have different binding subpopulations sizes in an infection. furthermore, measurement of icam-1 or cd36 binding sub-populations may practical to study the cytoadherence phenotypes of p. falciparum field isolates at the molecular level.

متن کاملα-Thalassemia Impairs the Cytoadherence of Plasmodium falciparum-Infected Erythrocytes
BACKGROUND α-Thalassemia results from decreased production of α-globin chains that make up part of hemoglobin tetramers (Hb; α(2)β(2)) and affects up to 50% of individuals in some regions of sub-Saharan Africa. Heterozygous (-α/αα) and homozygous (-α/-α) genotypes are associated with reduced risk of severe Plasmodium falciparum malaria, but the mechanism of this protection remains obscure. We h...
متن کاملImpaired cytoadherence of Plasmodium falciparum-infected erythrocytes containing sickle hemoglobin.
Sickle trait, the heterozygous state of normal hemoglobin A (HbA) and sickle hemoglobin S (HbS), confers protection against malaria in Africa. AS children infected with Plasmodium falciparum are less likely than AA children to suffer the symptoms or severe manifestations of malaria, and they often carry lower parasite densities than AA children. The mechanisms by which sickle trait might confer...
متن کاملParasite sequestration in Plasmodium falciparum malaria: spleen and antibody modulation of cytoadherence of infected erythrocytes.
Sequestration, the adherence of infected erythrocytes containing late developmental stages of the parasite (trophozoites and schizonts) to the endothelium of capillaries and venules, is characteristic of Plasmodium falciparum infections. We have studied two host factors, the spleen and antibody, that influence sequestration of P. falciparum in the squirrel monkey. Sequestration of trophozoite/s...
متن کاملEffects of sevuparin on rosette formation and cytoadherence of Plasmodium falciparum infected erythrocytes
In severe falciparum malaria cytoadherence of parasitised red blood cells (PRBCs) to vascular endothelium (causing sequestration) and to uninfected red cells (causing rosette formation) contribute to microcirculatory flow obstruction in vital organs. Heparin can reverse the underlying ligand-receptor interactions, but may increase the bleeding risks. As a heparin-derived polysaccharide, sevupar...
متن کاملSynergism of multiple adhesion molecules in mediating cytoadherence of Plasmodium falciparum-infected erythrocytes to microvascular endothelial cells under flow.
Plasmodium falciparum-infected erythrocytes (IRBCs) have been shown to interact with a number of endothelial adhesion molecules expressed on transfectants, on cell lines, and as immobilized purified receptor proteins under flow conditions. However, the experiments were designed in such a way that maximal numbers of adhesion molecules were provided as substratum. Whether the interactive events a...
